Purpose of assignment: The purpose of this activity is to give you practice performing the calculations involved with agarose gel electrophoresis assay including and buffer and sample preparation. The calulations performed in this activity are located at to the beginning wherase in the actual in-class activity they are scattered throughout. A big difference between this activity and the actual activity are the calculations that msut be performed. This activity is formatted very similarly to the actual in-class activity. Every time you run an agarose gel electrophoresis assay on your DNA samples, you should always include a DNA ladder as a refeerce sample next to your samples to help data interpreration. DNA ladders are referred to as a reference sample because it is used to help you understand your data. In this make-up lab assignment, you will perform the calculations necessary to prepare for and run an gel electrophoresis assay on a special sample called a DNA ladder.

For your education, the protocol the students are following for the in-class activity is colored in blue. However, this make-up assignment will focus on 1, preparing for agarose gel electrophoresis. That actual in-class activity asks each student to 1) prepare for agarose gel electrophoresis by performing the calculations necessary and 2) using the calculations and applying them to a protocol.
